Really, Who are they? Well, basically all we know are that they run the organization and they create new claymores, but how is it that simple? Let's bring up some points to who they really are.
Firstly, are they really human? I don't really see normal humans (in accordance to the manga and anime) controlling a whole fleet of half-humans half-yoma. Could it be they were actual successes of the first of the male claymore era?
Secondly (slight spoiler from the manga), have you noticed that they don't really age? Maybe they're awakened beings whom have maintained a human form like Priscilla which doesn't really age.
Anyways, discuss who or what they really are.

That's something I've wondered about. They don't age, many seem to have deformities, and they're all male.
Are they male Claymores who didn't go rogue, perhaps ones in a half-awakened state? But then who created them? Maybe they're people who experimented on themselves with yoma grafts, and then used a more powerful but unstable version to create the Claymores.
Which ties into something else I've wondered, which is what are yoma? I don't recall seeing a female yoma (true yoma, not awakened one). What are they, were do they come from, how do they reproduce?

There isn't much explained but then there's tons of theories and mysteries. Ya, I also wondered why there weren't any females who are yoma. Maybe it's cause that usually it's the guys-who-turn-out-to-be-monsters story type.

They COULD be the REAL masterminds behind the actual Claymores? Instead of the 'organization' maybe it's really them that are manipulating everything. 
They did not even explain how they actually put in parts of yoma into making claymores. How do you think this would end up? I am telling you this much, they are not human and very likely male claymores who did not awaken.

I think that they are claymore too if not those girls like clare miria or even the ranked 1 can kill all those people who create the organization and if they are not awakened beings nor a claymore the dwellers of the deep and even teresa when she was alive would destroy or kill the organization after all they know where is the hideout of the organization then why would the claymores are afraid of the organization especially when they are in the front of the leaders even the one who gives order or the one who gets the money price are scared to them shown when galatea was order to retrieve clare from missing. I think Yoma are not males nor female they only form to males/female because when they eat or get the humans memory they them self become that person just like the brother of raki and oh the real yoma not the claymore who awakened duh.

Like `Omnidevil said they could be male claymores, but that would be weird because males are more likely to awaken in a short time and even if they have such a strong mind to hold it back, at least some of them appear in periods about ten years or something, such an ammount of time without awakening might be a problem for most of the female claymores, so how a strong mind need a male one to prevent this about this time? I don't think it's possible.

Hmm...got some nice theories there... i really hope the mangaka will clear this up..if not itll jsut b one hole in this nice anime... which will be quite sad... =\
They must have eat the pills that makes your yoki seal for about a day and or maybe just a plain human with mysterious past. anyway we dont know if they died or how many generation those org. have replace each leader
Hmm I wondered... they don't age but is that for awakened beings too? I mean like priscilla looked always the same, does that mean that awakened beings can only die during battle?oO I'm pretty sure it's like that and the black suited guys have most of the awakened beings characteristics, but I'm sure they aren't like the typical ones. They seem to have a human mind and it would be weird anyway that they send claymores to defeat creatures of their own kind... Also they don't seem to hunt for human guts (or at least they were never shown being atracted by human flesh). But maybe that's just to confuse us... xD I'm sure the truth will be surprising, more or less....

Those guys are the worst,it's all about money and power to them.If the Org is all about protecting humans then why are they charging so high for each job?

An interesting question indeed. Quite a number of interesting theories shown in this thread as well. To add I guess I must say that the idea of the black-suits to be mere "male claymores" or "male awakened beings", i think is very unlikely. If I was the writer of Claymore, I'd say that to be able to control such an Organization which constitutes a hierarchy by power, these people must be on real different level. Maybe in a different level of power. If not in power, maybe a deep level of conspiracy enabling them even without power themselves to be able control beings with great power. A minor speculation for example, they must be the oldest of the yoma perhaps? Seeking to control the whole world through such a organization.
